Microscopy Shared Resource

The Microscopy Shared Resource provides core services to scientists engaged in a wide range of scientific endeavors. Among the types of services rendered are wide-field fluorescence microscopy, deconvolution microscopy, confocal microscopy including point-scanning and spinning disk, live cell imaging, super-resolution structured illumination microscopy, standard transmission electron microscopy, immunoelectron microscopy (enzymatic and gold labeling), DNA spreads and evaluation of subcellular fractionation procedures by negative staining or thin sectioning.
After training in the use of a particular microscope by Stephen Hearn, the Shared Resource Manager, investigators can sign up for microscope use. Scheduling of the major microscopes is done through on-line reservation calendars on a first come first served basis. Microscope images are captured using cooled CCD cameras and data sets can be directly downloaded to a shared server or to their own computers for preparing figures for presentation and/or publication.
In addition to providing users with access to state-of-the-art microscopes, the Shared Resource also provides technical assistance in preparing cells or tissues for immunofluorescence microscopy, transmission electron microscopy or immunoelectron microscopy. MICROSCOPES IN FACILITY:
- Zeiss Axioplan2i upright fluorescence microscope for examining fixed samples.
- Zeiss AxioImager.Z1 motorized upright fluorescence microscope for examining fixed samples.
- Zeiss Observer inverted fluorescence microscope equipped with a fully automated stage for image tiling/mosaics, time-lapse imaging of cultured cells, and brightfield imaging using phase-contrast or Nomarski optics.
- Zeiss LSM510 confocal laser scanning microscope equipped for 3-D imaging and photokenisis experiments, 3 laser lines (488, 568 and 633nm).
- Zeiss LSM710 confocal laser scanning microscope with live cell imaging capabilities (time-lapse and photokinesis) and fully automated stage for image tiling, 7 laser lines (405, 458, 488, 514, 561, 594, 633nm), and advanced spectral separation functions.
- Perkin-Elmer UltraVIEW VoX high speed spinning disk (Yokogawa® CSU-X1) laser confocal microscope with live cell imaging capability, time-lapse microscopy, photokinesis, multi-position image acquisition, 6 laser lines (405, 440, 488, 514, 561 and 640nm).
- Applied Precision DeltaVision wide-field fluorescence microscope for live cell imaging and deconvolution.
- Applied Precision OMX (Optical Microscope eXperimental) super-resolution microscope, 5 laser lines (405, 457, 488, 514, and 594nm).
- Hitachi H-7000 transmission electron microscope.
