DESCRIPTION

The DNA Sequencing Shared Resource provides access to high throughput next generation sequencing technologies and to full service DNA sequencing using fluorescence-based technology.

Next Generation Sequencing
The Woodbury genome center is home to three Illumina Genome Analyzers, nine Illumina HiSeq 2000, one Illumina Miseq, one Ion Torrent PGM and two Pacific Biosciences RS sequencers. This high-throughput sequencing technology offers a broad range of genetic analysis applications including genomic DNA sequencing/resequencing, siRNA, hybrid selection, chIP-seq, mRNA-seq, cDNA, PCR amplicons sequencing and digital gene expression. The HiSeqs can produce over 300 billion bases of sequence per in eleven days while the Miseq can yield over 1.5 billion bases in 27 hours. The Pacific Biosciences RS instruments can generate medium size sequences at high accuracy (300-600 bases) or long sequences (1 to 4 Kbases) at an 85% accuracy. To make this technology more widely accessible to the CSHL community, consulting is available on applications, experimental set-up and strategies for experimental design (contact Eric Antoniou).
Capillary Sequencing
The DNA Sequencing Shared Resource provides cost effective and rapid DNA sequence analysis using an ABI3730 instrument. The DNA Sequencing Facility offers full service fluorescent-based capillary DNA sequencing. Users can submit samples of PCR products, single and double stranded plasmids, BACs and cosmids. Facility staff set up reactions, run the samples and post final data on a shared drive for retrieval. The resource also provides consulting expertise for DNA preparation to improve the DNA sequencing results of the users. The on-site service provides convenience and a rapid turn-around time, facilitating the many projects that require DNA sequencing support.
